The exons 3~29 of mouse Adgrg2 gene that encodes the full-length protein was replaced by human ADGRG2 exons 3~28 in B-hADGRG2 mice.

ADGRG2 was detected in testis in both wild-type and homozygous B-hADGRG2 mice, as anti-ADGRG2 antibody was cross-reactive between human and mouse.
Protein expression analysis
Western blot analysis of ADGRG2 protein expression in homozygous B-hADGRG2 mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hADGRG2 mice (H/H), and then analyzed by western blot with anti-ADGRG2 antibody. 40 μg total proteins were loaded for western blotting analysis. ADGRG2 was detected in testis in both wild-type and homozygous B-hADGRG2 mice, as anti-ADGRG2 antibody was cross-reactive between human and mouse.
mRNA expression analysis
Strain specific analysis of ADGRG2 mRNA expression in wild-type C57BL/6 mice and B-hADGRG2 mice by RT-PCR. Testis RNA were isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hADGRG2 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human ADGRG2 primers. Mouse ADGRG2 mRNA were detectable only in wild-type C57BL/6 mice. Human ADGRG2 mRNA was detectable only in homozygous B-hADGRG2 mice but not in wild-type mice.
