As a negative regulator of the Wnt signalling pathway, APCDD1 acts upstream of beta-catenin and is involved in the regulation of normal cell development and homeostasis by inhibiting Wnt signalling through interactions with typical Wnt3a ligands and LRP5 receptors on the cell surface.

mRNA expression analysis: Mouse Apcdd1 was only detectable in wild-type C57BL/6JNifdc mice. Human APCDD1 was only detectable in homozygous B-hAPCDD1 mice.

Protein expression analysis: APCDD1 was detectable in wild-type C57BL/6JNifdc mice and homozygous B-hAPCDD1 mice, as anti-APCDD1 antibody was cross-reactive between mouse and human. APCDD1 was detectable in skin, subcutaneous fat, colon, heart, liver and kidney. APCDD1 expression is relatively low in skin and colon.
Protein expression analysis
Western blot analysis of APCDD1 protein expression in homozygous B-hAPCDD1 mice. Various tissue lysates were collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hAPCDD1 mice (H/H), and then analyzed by western blot with anti-APCDD1 antibody (Novus Biologicals, NB110-92756SS). 40 μg total proteins were loaded for western blotting analysis. APCDD1 was detected in skin, subcutaneous fat, colon, heart, liver and kidney.
mRNA expression analysis
Strain specific analysis of APCDD1 mRNA expression in wild-type C57BL/6JNifdc mice and B-hAPCDD1 mice by RT-PCR. Kidney RNA was isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hAPCDD1 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human APCDD1 primers. Mouse Apcdd1 mRNA was only detectable in wild-type C57BL/6JNifdc mice. Human APCDD1 mRNA was only detectable in homozygous B-hAPCDD1 mice.
