Description

• SIGLEC6 is an inhibitory receptor selectively expressed on human mast cells and is a member of a family of receptors that bind to sialoglycan.
• The full coding CDS, the promoter, 5’UTR and 3’UTR sequences of human SIGLEC6 gene are inserted into mouse Hipp11 (H11) locus in B-hSIGLEC6 mice(C).
• Human SIGLEC6 mRNA  was exclusively detectable in homozygous B-hSIGLEC6 mice(C).
• Human SIGLEC6 protein was detectable on mast cells in the peritoneal lavage fluid, B cells, NK cells, DCs, neutrophils, monocytes and macrophages in the blood of homozygous B-hSIGLEC6 mice(C).
• Application: This product is used for pharmacodynamics and safety evaluation of  drugs.

mRNA expression analysis

Species specific analysis of SIGLEC6 gene expression in wild-type BALB/c mice and homozygous humanized B-hSIGLEC6 mice(C) by RT-PCR. Spleen was collected from wild-type BALB/c mice (+/+) and homozygous B-hSIGLEC6 mice(C) (H/H). Human SIGLEC6 mRNA was detectable only in homozygous B-hSIGLEC6 mice(C), but not in wild-type BALB/c mice.

Protein expression analysis in mast cells

Strain specific SIGLEC6 expression analysis in homozygous B-hSIGLEC6 mice(C) by flow cytometry. Peritoneal lavage fluid was collected from wild-type BALB/c mice (+/+) and homozygous B-hSIGLEC6 mice(C) (H/H), and protein expression was analyzed with anti-human SIGLEC6 antibody (BD, 747915) by flow cytometry. hSIGLEC6 was only detectable in mast cells of homozygous B-hSIGLEC6 mice but not in wild-type BALB/c mice.
