Description

Coronin 1A is found exclusively in hematopoetic lineage cells such as lymphocytes, macrophages and neutrophils, and is required for appropriate immune regulation in mice and humans.

The exons 2-11 of mouse Coro1a gene that encode the whole molecule (ATG to STOP codon) were replaced by human counterparts in B-hCORO1A mice. The promoter, 5’UTR and 3’UTR region of the mouse gene are retained. The human CORO1A expression is driven by endogenous mouse Coro1a promoter, while mouse Coro1a gene transcription and translation will be disrupted.

CORO1A protein was detected in lung (weak expression), spleen, thymus, bone marrow, and lymph nodes of homozygous B-hCORO1A mice.

Protein expression analysis
Strain specific CORO1A expression analysis in homozygous B-hCORO1A mice by western blot. Lung, spleen, thymus, bone marrow, and lymph nodes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCORO1A mice (H/H), and analyzed by western blot with anti-CORO1A antibody (Proteintech, 17760-1-AP). m/hCORO1A was detectable in lung, spleen, lymph nodes, bone marrow, and thymus of wild-type C57BL/6 mice (+/+) and homozygous B-hCORO1A mice.
mRNA expression analysis
Strain specific analysis of Coro1a mRNA expression in wild-type C57BL/6 mice and B-hCORO1A mice by RT-PCR. Spleen RNA was isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hCORO1A mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CORO1A primers. Mouse Coro1a mRNA was detectable only in wild-type C57BL/6 mice. Human CORO1A mRNA was detectable only in homozygous B-hCORO1A mice but not in wild-type mice.
