Description

  • CD42b is a 145 kD glycoprotein known as GpIbα. It is covalently bonded to CD42c to form GPIb. 
  • CD42b is expressed on platelets and megakaryocytes. CD42b/c heterodimer forms a complex with CD42a and d and acts as the receptor for von Willibrand factor and thrombin.

Targeting strategy

Gene targeting strategy for B-hGP1BA mice. The exon 2 of mouse Gp1ba gene that encode extracellular domain is replaced by human counterparts in B-hGP1BA mice. The promoter and 5’UTR and 3’UTR region, transmembrane and cytoplasmic domain of the mouse gene are retained. The chimeric GP1BA expression is driven by endogenous mouse Gp1ba promoter, while mouse Gp1ba gene transcription and translation will be disrupted. 

Protein expression analysis in platelet and bone marrow

Strain specific GP1RA expression analysis in homozygous B-hGP1BA mice by flow cytometry. Platelet and bone marrow was collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hGP1BA mice (H/H), and analyzed by flow cytometry with species-specific anti-human GP1BA/CD42b antibody (BD, 551061). Human GP1BA/CD42b was exclusively detectable in homozygous B-hGP1BA mice but not in wild-type mice.

mRNA expression analysis

Strain specific analysis of GP1BA mRNA expression in wild-type C57BL/6JNifdc mice and homozygous B-hGP1BA mice by RT-PCR. Spleen RNA were isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hGP1BA mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse and human primers. Mouse Gp1ba mRNA were detectable only in wild-type mice. Human GP1BA mRNA were detectable only in B-hGP1BA mice but not in wild-type mice.
