Description

Siglec-1, also known as CD169 and Sialoadhesin (Sn), is a type I lectin containing 17 immunoglobulin (Ig) domains (one variable domain and 16 constant domains), which involved in macrophage binding to different cell types such as granulocytes, monocytes, NK, B, and T cells.

The exons 2-19 of mouse Siglec1 gene that encode extracellular domain and part of transmembrane domain are replaced by human counterparts in B-hSIGLEC1 mice. The genomic region of mouse Siglec1 gene that encodes signal peptide and cytoplasmic portion is retained. The promoter, 5’UTR and 3’UTR region of the mouse gene are also retained. The chimeric SIGLEC1 expression is driven by endogenous mouse Siglec1 promoter, while mouse Siglec1 gene transcription and translation will be disrupted.

Strain specific SIGLEC1 expression analysis in homozygous B-hSIGLEC1 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hSIGLEC1 mice (H/H), and protein expression was analyzed with anti-mouse SIGLEC1 antibody (Biolegend, 142403) and anti-human SIGLEC1 antibody (Biolegend, 346007) by flow cytometry. mSIGLEC1 was detectable in NK cells of wild-type C57BL/6JNifdc mice, but not T cells and B cells. hSIGLEC1 was detectable in NK cells of homozygous B-hSIGLEC1 mice, but not T cells and B cells.
