    Novel IL4/IL4RA double humanized mouse models for in vivo efficacy evaluation of human IL4 and human IL4RA antibodies in asthma models

    Recently, the key role played by interleukin (IL)4/13 and IL4 receptor in the pathophysiology of asthma, atopic dermatitis (AD), and other auto-immune diseases has been widely accepted, and the pleiotropic cytokines are now considered to be suitable therapeutic targets. The monoclonal antibody, Dupilumab, is a very promising therapeutic agent because of its ability to inhibit the biological effects of both IL4 and IL13 and prevent the interaction of IL4/IL13 with the α-subunit of the IL-4 receptor complex (IL4RA). However, human IL4 antibodies are specific to human IL4RA and can not cross-react with mouse endogenous IL4RA, and mouse IL4 can not recognize human IL4RA. To solve this problem, we generated IL4/IL4RA double humanized mouse model for in vivo efficacy evaluation of IL4 and IL4RA antibodies.
