      mRNA expression analysis
      Strain specific analysis of APOE2 mRNA expression in wild-type C57BL/6J mice and B-hAPOE2 mice by RT-PCR. Brain RNA was isolated from wild-type C57BL/6J mice (+/+) and homozygous B-hAPOE2 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human APOE primers. Mouse Apoe mRNA was detectable only in wild-type C57BL/6J mice. Human APOE2 mRNA was detectable only in homozygous B-hAPOE2 mice but not in wild-type mice.
      Protein expression analysis

      Strain specific APOE2 expression analysis in homozygous B-hAPOE2 mice by ELISA. Serum was collected from wild-type mice C57BL/6 mice (+/+) and homozygous B-hAPOE2 mice (H/H), and analyzed by ELISA with species-specific APOE ELISA kit (Abcam, ab233623; Abcam, ab215086). Mouse APOE was detectable in wild-type mice. Human APOE2 was exclusively detectable in homozygous B-hAPOE2 mice but not in wild-type mice.
