• S1PR4 (Sphingosine-1-phosphate receptor 4) is a member of the G protein-coupled receptor (GPCR) family. It primarily binds to sphingosine-1-phosphate (S1P) and is involved in regulating various physiological and pathological processes. It is predominantly expressed in the immune system.
• The exon 1 of human S1PR4 gene that encodes the full-length protein and 3’UTR was inserted into the exon 1 of mouse S1pr4 in B-hS1PR4 mice. The promoter and 5’UTR region of mouse gene are retained. The human S1PR4 expression is driven by endogenous mouse S1pr4 promoter.
• Human S1PR4 mRNA  was exclusively detectable in homozygous B-hS1PR4 mice.
• S1PR4 protein was detected in spleen and thymus of wild-type C57BL/6N mice and homozygous B-hS1PR4 mice, as the antibody can cross-recognize both human and mouse S1PR4.
• Application: This product is used for pharmacodynamics and safety evaluation of  drugs.

Species specific analysis of S1PR4 gene expression in wild-type C57BL/6N mice and homozygous humanized B-hS1PR4 mice by RT-PCR. Spleen was collected from wild-type C57BL/6N mice (+/+) and homozygous B-hS1PR4 mice (H/H). Mouse S1pr4 mRNA was only detectable in wild-type C57BL/6N mice. Human S1PR4 mRNA was exclusively detectable in homozygous B-hS1PR4 mice but not in wild-type C57BL/6N mice.

Western blot analysis of S1RP4 protein expression in homozygous B-hS1RP4 mice. Various tissue lysates were collected from wild-type C57BL/6N mice (+/+) and homozygous B-hS1RP4 mice (H/H), and then analyzed by western blot with cross reactive anti-S1RP4 antibody (Invitrogen, PA5-102054). 40 μg total proteins were loaded for western blotting analysis. S1RP4 was detected in spleen and thymus of wild-type C57BL/6N mice and homozygous B-hS1PR4 mice.
