Description
- ADAMTS13 is a metalloproteinase encoded by the ADAMTS13 gene. Its primary function is to cleave von Willebrand factor (VWF). A deficiency in ADAMTS13 can result in thrombotic thrombocytopenic purpura (TTP). This dysfunction highlights the enzyme's critical role in preventing microthrombosis by modulating VWF multimer size.
- The exons 1-23 of mouse Adamts13 gene were knocked out in B-Adamts13 KO mice.
- Mouse Adamts13 mRNA were detectable only in wild-type C57BL/6 mice but not in homozygous B-Adamts13 KO mice.
- Application: This product is used for pharmacodynamics and safety evaluation of thrombotic thrombocytopenic purpura (TTP).
Targeting strategy
Gene targeting strategy for B-Adamts13 KO mice. The exons 1-23 of mouse Adamts13 gene were knocked out in B-Adamts13 KO mice.
mRNA expression analysis
Strain specific analysis of Adamts13 mRNA expression in wild-type C57BL/6 mice and B-Adamts13 KO mice by RT-PCR. Liver RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-Adamts13 KO mice (-/-), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse Adamts13 primers. Mouse Adamts13 mRNA were detectable only in wild-type C57BL/6 mice but not in homozygous B-Adamts13 KO mice.
