      Gene targeting strategy for B-hGPRC5D mice. The exons 2-4 of mouse Gprc5d gene that encode the full-length protein were replaced by human GPRC5D exons 1-3 in B-hGPRC5D mice.

      mRNA expression analysis
      Strain specific analysis of GPRC5D gene expression in wild-type C57BL/6 mice and B-hGPRC5D mice by RT-PCR. Mouse Gprc5d mRNA was detectable only in testis of wild-type C57BL/6 mice (+/+). Human GPRC5D mRNA was detectable only in homozygous B-hGPRC5D mice (H/H), but not in wild-type mice.
      Protein expression analysis

      Immunohistochemical (IHC) analysis of GPRC5D expression in homozygous B-hGPRC5D mice. The heart, liver, spleen, lung, kidney, brain, tongue, skin and salivary glands were collected from C57BL/6JNifdc(+/+) (female, 7-week-old) and homozygous B-hGPRC5D mice(H/H) (female, 7-week-old), analyzed by IHC with anti-human GPRC5D (abcam, ab315808). GPRC5D was detectable in tongue and skin of homozygous B-hGPRC5D mice. The arrow indicates tissue cells with positive GPRC5D staining (brown). “+” indicates that the tissue is positive and “-” indicates that the tissue is negative. Note: This antibody shows non-specific staining in muscle tissue.
